Neuropathology: Many Paths Lead to Hereditary Spastic Paraplegia
نویسندگان
چکیده
Studies with animal models are providing new insights into the pathology of hereditary spastic paraplegia, particularly how mutations in multiple, converging pathways can lead to this family of neuropathies.

Clinical Neuropathology image 1-2015: Crystal-storing histiocytosis of the central nervous system
Crystalloid inclusions are rarely encountered within the central nervous system (CNS). Aside from the more common Hirano bodies and Rosenthal fibers, which occur within neurons and astrocytic processes, both being composed of cytoskeletal proteins, eosinophilic crystalloid inclusions have been recently described within oligodendrocytes in a case of adult-onset, complicated form of hereditary sp...
متن کاملStrümpell's familial spastic paraplegia: genetics and neuropathology.
Uncomplicated Strümpell's disease (Strümpell's familial spastic paraplegia) with a dominant mode of inheritance is recorded in six families. The neuropathological findings in two cases from these families are given, bringing the total of similar histologically documented reports in the literature to 11. It is concluded that, although exact classification and identification of the many different...
متن کاملSPATACSIN mutations cause autosomal recessive juvenile amyotrophic lateral sclerosis
The mutation of the spatacsin gene is the single most common cause of autosomal recessive hereditary spastic paraplegia with thin corpus callosum. Common clinical, pathological and genetic features between amyotrophic lateral sclerosis and hereditary spastic paraplegia motivated us to investigate 25 families with autosomal recessive juvenile amyotrophic lateral sclerosis and long-term survival ...
متن کاملMultigeneration family with dominant SPG30 hereditary spastic paraplegia
Autosomal recessive KIF1A missense mutations cause hereditary spastic paraplegia (HSP) type SPG30, while recessive truncations lead to sensory and autonomic neuropathy (HSN2C) and many de novo missense mutations are associated with cognitive impairment. Here, we describe family members across three generations with pure HSP. A heterozygous p.Ser69Leu KIF1A mutation segregates with those afflict...
متن کاملPathogenesis of Autosomal Dominant Hereditary Spastic Paraplegia (SPG6) Revealed by a Rat Model
Hereditary spastic paraplegias (HSPs) are characterized by progressive spasticity and weakness in the lower extremities that result from length-dependent central to peripheral axonal degeneration. Mutations in the non-imprinted Prader-Willi/Angelman syndrome locus 1 (NIPA1) transmembrane protein cause an autosomal dominant form of HSP (SPG6). Here, we report that transgenic (Tg) rats expressing...
